Nested Pcr

Nested PCR is a molecular amplification method that uses two successive rounds of polymerase chain reaction to increase the specificity and sensitivity of target DNA detection. The first primer pair amplifies a broader region, while a second primer pair binds within that initial product and amplifies an internal sequence, reducing amplification from nonspecific products. In immunology and infection research, nested PCR can detect low-abundance pathogen genomes or immune-related genetic targets in complex clinical and environmental samples. Its improved target discrimination supports studies of infection, pathogen identification, and molecular diagnosis when conventional PCR produces weak or ambiguous results.

This video describes nested polymerase chain reaction, a technique that consists of two sequential PCR amplification processes using two primer sets. The first set of primers is intended to anneal to sequences upstream of the second set, resulting in selective amplification of specific gene sequences. This PCR is more sensitive and specific than a normal PCR and is widely used as a detection technique for various diseases.

Detecting the Lyme Disease Spirochete, Borrelia Burgdorferi, in Ticks Using Nested PCR

Nested PCR is a sensitive, specific, and straightforward technique that can be applied to tick DNA extracts to probe for Borrelia burgdorferi, the causative agent of Lyme disease. The initial PCR experiment uses gene-specific primers to generate long amplicons, which then become templates for a subsequent reaction using internal primers.

Nest Building as an Indicator of Health and Welfare in Laboratory Mice

We demonstrate the utility of nest building behavior in laboratory mice as an indicator of welfare. Nest scoring is a sensitive technique that is altered by temperature, illness, and aggression. The time to integrate into nest test (TINT) is a simple cage-side assessment that can detect postoperative pain.

Assessing Burrowing, Nest Construction, and Hoarding in Mice

Burrowing, nesting, and hoarding are species-typical activities that mice readily perform in the laboratory. This article describes how they can be easily and cheaply assessed. These protocols are extremely sensitive to mouse strain, brain lesions and diseases. Moreover they constitute “environmental enrichment” for the mice, and embody the “Refinement” aspect of the “3 Rs”18.

A Push-pull Protocol to Reduce Colonization of Bird Nest Boxes by Honey Bees

Preventing colonization of bird nest boxes by invasive Africanized honey bees is important for conservation efforts of nest-site limited birds. We provide an integrated pest management approach to "push" bees away from nest boxes with a repellant insecticide, permethrin, and "pull" them toward pheromone baited swarm traps.
