Quanti-blue Substrate

Quanti-Blue substrate is a colorimetric reagent used to detect secreted alkaline phosphatase (SEAP) activity in cell culture samples, providing a convenient readout for reporter assays in immunology and infection research. When SEAP is released into the culture medium, it enzymatically converts components of the Quanti-Blue reagent into a blue-purple product, with color intensity reflecting reporter activity and measured by visual inspection or absorbance. Researchers use this assay to monitor signaling pathways such as NF-κB and NFAT, assess innate immune responses to microbial stimuli, and evaluate cytokine or pattern-recognition receptor activity. Its nonradioactive format supports rapid, scalable screening of immune activation and inhibition.

Exfoliation of Egyptian Blue and Han Blue, Two Alkali Earth Copper Silicate-based Pigments

The preparation and exfoliation of CaCuSi4O10 and BaCuSi4O10 are described. Upon stirring in hot water, CaCuSi4O10 spontaneously exfoliates into monolayers, whereas BaCuSi4O10 requires ultrasonication in organic solvents. Near infrared (NIR) imaging illustrates the NIR emission properties of these materials, and aqueous dispersions of these nanomaterials are useful for solution processing.

CRISPR/Cas systems mediate adaptive immunity in Bacteria and Archaea. Many Cas proteins are proposed to act as endoribonucleases acting on crRNA precursors of varying length. Here we illustrate three different approaches to generate pre-crRNA substrates for the biochemical analysis of Cas endonuclease activity.

Measurement of Carbon Dioxide Production from Radiolabeled Substrates in Drosophila melanogaster

This paper describes a method for the measurement of fuel oxidation in Drosophila melanogaster in which trace amounts of specific radiolabeled metabolic substrates are fed to flies. The exhaled radiolabeled CO2 that is a produced from fuel oxidation is collected and measured.
