Serially Truncated Proteins

Serially truncated proteins are engineered protein variants that progressively lack defined segments, allowing researchers to map regions responsible for structure, binding, or biological activity. By creating a systematic series of deletion constructs and comparing their expression, stability, antigen recognition, or function with the full-length protein, investigators can identify essential domains and boundaries. In immunology and infection research, this approach helps localize antibody and T-cell epitopes, characterize pathogen virulence factors, and determine host-interaction regions in microbial proteins. The resulting maps support antigen design, vaccine development, diagnostic assay optimization, and the study of molecular mechanisms underlying infection and immune recognition.

Truncation in Survival Analysis

Truncation in survival analysis refers to the exclusion of individuals or events from the dataset based on specific criteria related to the time of the event. This exclusion can happen in two primary forms: left truncation and right truncation. Left truncation occurs when individuals who experienced the event of interest before a certain time are not included in the study. This is often due to a "delayed entry" into the study where only those who survive until a certain entry point are observed.

Preparation and Delivery of Protein Microcrystals in Lipidic Cubic Phase for Serial Femtosecond Crystallography

We describe procedures for the preparation and delivery of membrane protein microcrystals in lipidic cubic phase for serial crystallography at X-ray free-electron lasers and synchrotron sources. These protocols can also be applied for incorporation and delivery of soluble protein microcrystals, leading to substantially reduced sample consumption compared to liquid injection.
