Simultaneous Quantification

Simultaneous quantification is the measurement of multiple analytes in a single sample during the same analytical process, improving efficiency and enabling direct comparison of biological signals. In immunology and infection research, multiplex assays use distinct detection reagents, signal labels, or assay regions to identify and quantify targets such as cytokines, antibodies, antigens, or pathogen markers, with calibration standards supporting accurate concentration estimates. This approach helps characterize coordinated immune responses, distinguish infection-associated signatures, and assess treatment effects while conserving limited samples. Its ability to capture complex interactions in parallel supports biomarker discovery, disease monitoring, and more comprehensive experimental analysis.

Duplex Digital PCR for Simultaneous Quantification of Dual Genetic Markers

2025

In this video, we demonstrate the duplex digital PCR (ddPCR) technique — a modification of the traditional PCR technique that is useful in detecting two different genetic markers simultaneously. A single PCR reaction is partitioned into nanoliter-sized emulsified droplets that are independently amplified, and the detection of differently colored fluorescence amplification signals from the fraction of droplets is used to compute the initial concentration of the target sequences.

This workflow describes the performance of time- and cost-efficient enrichment of multiple protein post-translational modifications (PTMs) simultaneously for quantitative global proteomic analysis. The protocol utilizes peptide-level PTM enrichment with multiple conjugated antibodies, followed by data-independent acquisition mass spectrometry analysis to gain biological insights into PTM crosstalk.
