Vasoactive Drugs

Vasoactive drugs are medications that alter vascular tone and, consequently, blood pressure, blood flow, and tissue perfusion. Vasoconstrictors such as norepinephrine activate adrenergic receptors on vascular smooth muscle to increase systemic vascular resistance, while vasodilators relax these vessels and reduce pressure or cardiac workload. In immunology and infection, these agents are especially important during sepsis and septic shock, when inflammatory mediators can cause widespread vasodilation, capillary leakage, and inadequate organ perfusion. By supporting circulation and guiding hemodynamic management, vasoactive drugs help stabilize patients while antimicrobial therapy and treatment of the underlying infection address its cause.

The small intestine is frequently exposed to toxins that can influence blood flow and negatively impact nutrient absorption. Using a multimyograph and mesenteric artery and vein isolates, compounds or toxins of interest can be screened for vasoactivity.

Pressure myography is used to assess vasoactivity of small arteries that develop sustained constriction when pressurized. This manuscript provides a detailed protocol to assess in isolated segments of small mesenteric arteries from rats, vasoactivity and the effect of intraluminal pressure on vascular diameter.

A validated computerized battery of neuropsychological and neurophysiological tests is used to study pharmacodynamic effects on the central nervous system of newly developed drugs in early phase development. To demonstrate the test battery, the acute effects of mecamylamine and the reversal of these effects by two agonist drugs are described.

Drug–drug interactions can precipitate toxicity through multiple mechanisms.
