Calcium Signal Recording

Calcium signal recording is the measurement of changes in intracellular calcium concentration over time to study how cells communicate, respond to stimuli, and regulate physiological activity. In a typical approach, fluorescent calcium indicators bind calcium or change their fluorescence when calcium levels shift, allowing microscopy or specialized detectors to track calcium influx, release from intracellular stores, and recovery toward baseline. In medicine, these recordings help characterize neuronal signaling, muscle contraction, endocrine secretion, and cardiac activity, while also supporting research into disease mechanisms and drug responses. Quantifying signal amplitude, timing, and spatial patterns can reveal abnormal cellular function and guide the development of diagnostic and therapeutic strategies.

This protocol outlines a simple method for analyzing calcium signals in plants generated by feeding hemipteran insects, such as aphids. Arabidopsis thaliana transformed with the GFP calcium biosensor GCaMP3 allow for the real-time in vivo imaging of calcium dynamics with a high temporal and spatial resolution.

The fruit fly, Drosophila melanogaster, extends its proboscis for feeding, responding to a sugar stimulus from its proboscis or tarsus. I have combined observations of the proboscis extension response (PER) with a calcium imaging technique, allowing us to monitor the activity of neurons in the brain, simultaneously with behavioral observation.

Procedures are described to perform simultaneous recordings of membrane potential or current and changes of intracellular calcium concentration. Suprachiasmatic nucleus neurons are filled with the calcium indicator bis-fura-2 using a patch clamp electrode in the whole cell patch clamp configuration.
