Cholesterol Anchoring

Cholesterol anchoring is a lipid-based strategy that attaches a cholesterol group to a molecule, enabling it to associate with cell membranes and other lipid structures. The hydrophobic cholesterol moiety inserts into the membrane’s lipid bilayer while the linked protein, peptide, nucleic acid, or drug remains exposed to the aqueous environment, creating a stable membrane tether whose behavior depends on linker chemistry and membrane composition. In medicine, cholesterol anchoring supports targeted delivery, improves cellular uptake and retention of therapeutic molecules, and helps organize biomolecules for studying membrane signaling, trafficking, and interactions. This approach also informs the design of lipid-based therapeutics and biomaterials.
