Drug-naive Patients

Drug-naive patients are individuals who have not previously received a particular medication or therapeutic drug class, making their baseline response especially valuable in medicine and clinical research. Because prior exposure has not altered drug sensitivity, tolerance, resistance, immune status, or treatment-related biomarkers, clinicians and researchers can assess pharmacologic effects with fewer confounding influences. Drug-naive populations are therefore important in clinical trials, treatment guidelines, and studies of initial therapy, where outcomes such as efficacy, adverse effects, and disease response can be evaluated before subsequent treatments complicate interpretation. The term is context-dependent and does not necessarily mean that a patient has never taken any medication.

Naïve CD4+ T cells polarize to various subsets depending on the environment at the time of activation. The differentiation of naïve CD4+ T cells to various effector subsets can be achieved in vitro through the addition of T cell receptor stimuli and specific cytokine signals.

We describe a method for generating regulatory, memory and naïve T cells from a single human blood donor. Polarized Tregs can be then compared to other subsets in a variety of genetic and functional applications with genetic homogeneity, including a suppression assay also detailed here.

Drug Dosing: Obese Patients

2025

In the United States, obesity is a prominent concern. It is linked to heightened mortality rates due to increased occurrences of conditions such as hypertension, atherosclerosis, coronary artery disease, and diabetes compared to nonobese individuals. A patient is classified as obese if their actual body weight surpasses the ideal or desirable body weight by 20%, based on Metropolitan Life Insurance Company data. Ideal body weights consider average weights and heights for males and females...

With effector functions distinct from other T cell subsets, Th17 cells have been centrally implicated in inflammatory autoimmunity. This in vitro Th17 differentiation protocol provides a means to determine whether naïve CD4+ T lymphocytes can differentiate into Th17 cells, and to further examine their role in autoimmunity and host response.

Drug Dosing: Geriatric Patients

Elderly individuals encompass a diverse population with varying degrees of age-related physiological changes. Defining the elderly presents challenges, as the geriatric population is often arbitrarily categorized as individuals older than 65. However, many individuals in this group lead active and healthy lives, with an increasing number surpassing 85 years and falling into the older elderly category. Physiological changes associated with aging impact performance capacity and homeostatic...
