Murine Vascular Model

Murine vascular models are experimental systems that use mice to investigate blood vessels, vascular function, and disease-related changes in a living organism. Researchers examine how genetic alterations, disease stimuli, or therapeutic interventions affect endothelial cells, vascular smooth muscle, vessel permeability, blood flow, and remodeling through physiological, imaging, and molecular analyses. These models help clarify mechanisms underlying cardiovascular disease, inflammation, tumor angiogenesis, and impaired tissue perfusion while providing a practical platform for evaluating candidate drugs and procedures. Their similarities and differences with human vascular biology also guide interpretation of preclinical findings and the design of clinically relevant studies.

The purpose of this protocol is to demonstrate the isolation and culture techniques of murine primary vascular smooth muscle cells (VSMCs) from the coronary circulation. Once VSMCs have been isolated, they can be used for many standard culture techniques.

We present a protocol to isolate adipose tissue-derived microvascular fragments that represent promising vascularization units. They can be rapidly isolated, do not require in vitro processing and, thus, may be used for one-step prevascularization in different fields of tissue engineering.
