Axonal Degeneration

Axonal degeneration is the progressive breakdown of an axon, the long neuronal projection that transmits signals, and it is a major feature of nervous system injury and disease. After axonal damage or loss of essential transport, disrupted energy metabolism and calcium influx activate proteases such as calpain, causing cytoskeletal disassembly, axon fragmentation, and eventual clearance by glial cells. In neuroscience, studying processes such as Wallerian degeneration helps explain how peripheral nerve injuries, neurodegenerative disorders, and toxic exposures impair neural connectivity. These mechanisms also guide the development of biomarkers and treatments designed to preserve axons, limit secondary damage, or promote nerve regeneration.

Here we present a protocol using nerve conduction studies and ultrasound to screen potential axonal degeneration associated with carpal tunnel syndrome. The criteria for differentiation are established. Compared to conventional approaches, this method is noninvasive, convenient, and efficient, with an overall satisfactory accuracy, sensitivity, and specificity.
