Durotomy Procedure

A durotomy procedure is a controlled surgical incision through the dura mater, the tough outer membrane surrounding the brain and spinal cord, to provide access to underlying neural tissues. Surgeons create the opening with microsurgical instruments while protecting the cortex, spinal cord, blood vessels, and cerebrospinal fluid pathways, then close the dura watertight to restore protection and reduce cerebrospinal fluid leakage. In neuroscience, durotomy enables treatment of intracranial and spinal conditions, including tumor removal, vascular lesion repair, decompression, and access for experimental procedures. Its careful execution supports effective intervention while limiting tissue injury and postoperative complications.

This video and protocol demonstrate how to implant a glass-covered cranial window in rodents. Here we present a protocol to visualize spatial correlation of calcitonin gene-related peptide (CGRP)-immunoreactive nerve fibers and blood vessels in the cranial dura mater using immunofluorescence and fluorescent histochemistry with CGRP and phalloidin, respectively. In addition, the origin of these nerve fibers was retrograde traced with a fluorescent neural tracer.
