Focused Ion Beam

Focused ion beam (FIB) is a precision microscopy and materials-processing technique that uses a finely focused beam of charged ions to image, mill, or modify a sample at nanometer-scale resolution. In operation, accelerated ions strike the surface, generating secondary electrons for imaging while sputtering material away; controlled beam current, dwell time, and stage movement determine the depth and shape of the cut. In neuroscience, FIB combined with scanning electron microscopy can serially section resin-embedded or frozen biological samples, enabling three-dimensional reconstruction of neurons, synapses, axons, and organelles. These datasets help link ultrastructure to neural function and disease, while targeted milling can prepare regions for correlative light and electron microscopy.

Cryo Electron Microscopes, either Scanning (SEM) or Transmission (TEM), are widely used for characterization of biological samples or other materials with a high water content1. We describe the approaches for the device fabrication and electrical characterization of molybdenum diselenide (MoSe2) layer semiconductor nanostructures with different thicknesses. In addition, the fabrication of ohmic contacts for MoSe2-layer nanocrystals by the focused-ion beam deposition method using platinum (Pt) as a contact metal is described.
