Functional Ultrasound

Functional ultrasound is a neuroimaging technique that maps brain activity by detecting task-related changes in cerebral blood flow, offering high spatial and temporal resolution without ionizing radiation. It uses ultrafast ultrasound pulses and Doppler processing to measure blood-volume or blood-flow variations associated with neurovascular coupling, often through an acoustic window in the skull or during surgery. In neuroscience, functional ultrasound supports studies of brain connectivity, sensory processing, behavior, and disease-related vascular changes in animal models and, increasingly, clinical settings. Its portability and sensitivity also make it valuable for monitoring brain function when conventional imaging is impractical.

Diagnostic ultrasound imaging has proven to be effective in diagnosing various respiratory diseases in human and animal subjects. We demonstrate a comprehensive ultrasound protocol utilized by Dr. Zuo's lab to analyze diaphragm kinetics specifically in mouse models. This is also a non-invasive research technique which can provide quantitative information on mouse respiratory muscle function.

Functional Imaging of Cerebral Blood Flow Using Transcranial Doppler Ultrasound

Source: Hage, B. D. et. al., Functional Transcranial Doppler Ultrasound for Monitoring Cerebral Blood Flow. J. Vis. Exp. (2021) This video demonstrates the use of Doppler ultrasound as a functional imaging tool to measure cerebral blood flow velocity, or CBFV in real time. It highlights how frequency shifts in the middle cerebral artery during tasks like breath-holding reveal changes in CBFV, offering insights into brain activity and vascular dynamics.

The flow mediated dilation (FMD) test is the most commonly utilized, non-invasive, ultrasound assessment of endothelial function in humans. Although the FMD test has been related with the prediction of future cardiovascular disease and events, it is a physiological assessment with many inherent confounding factors that need to be considered.

We describe a new ultrasound-based vector tissue Doppler imaging technique to measure muscle contraction velocity, strain and strain rate with sub-millisecond temporal resolution during dynamic activities. This approach provides complementary measurements of dynamic muscle function and could lead to a better understanding of mechanisms underlying musculoskeletal disorders.
