Neuronal Ensemble Recording

Neuronal ensemble recording is the simultaneous measurement of activity from multiple neurons to reveal how groups of cells represent information and coordinate behavior. In neuroscience, electrodes or optical sensors detect action potentials or changes in intracellular calcium as neurons respond to sensory input, movement, or task-related events. Analyzing the timing and patterns of these signals can identify population codes, functional connectivity, and coordinated network dynamics that are not apparent from single-neuron recordings. The approach supports research on learning, memory, perception, and motor control, while also helping investigators understand how abnormal ensemble activity contributes to neurological disorders.

Recording Large-scale Neuronal Ensembles with Silicon Probes in the Anesthetized Rat

Extracellular recordings of neuronal activity using silicon probes in the anesthetized rat will be described. This technique allows information to be obtained across multiple brain areas from more than 100 neurons simultaneously. It provides information with single cell resolution about neuronal ensembles dynamics in multiple local circuits.

Here, we employ HD-MEA to delve into computational dynamics of large-scale neuronal ensembles, particularly in hippocampal, olfactory bulb circuits, and human neuronal networks. Capturing spatiotemporal activity, combined with computational tools, provides insights into neuronal ensemble complexity. The method enhances understanding of brain functions, potentially identifying biomarkers and treatments for neurological disorders.

This video demonstrates establishing a neuron-astrocyte co-culture for recording neuronal network activity. Upon establishing the co-culture of motor neurons and astrocytes on a multi-electrode array (MEA) plate, the synchronous rhythmic electrical activity of the neuronal network was recorded extracellularly using the MEA.

This protocol describes in vivo GCaMP calcium imaging of intact trigeminal ganglion (TG) neurons. We present methods for the construction of electrodes to simultaneously record extracellular neural activity and release multiple neuroactive substances at the vicinity of the recording sites in awake mice. This technique allows the detailed analysis of putative local synaptic inputs to the neuron of interest.
