Spinal Neurodegeneration Models

Spinal neurodegeneration models are experimental systems that reproduce key features of progressive neuronal loss and circuit dysfunction in the spinal cord, enabling researchers to study disease mechanisms and potential treatments. These models may use cultured neurons, organoids, genetically modified organisms, or injury-based paradigms to examine processes such as axonal degeneration, motor neuron death, glial activation, and impaired neuroinflammation or repair. In neuroscience, they help connect cellular and molecular changes with motor deficits, support biomarker identification, and provide platforms for testing drugs, gene therapies, and regenerative strategies. Careful model selection is essential because each system captures different aspects of human spinal disease.

Establishing a Model of Zika Virus-Induced Neurodegeneration in an Adult Mouse

The video demonstrates intracerebral injection of Zika virus suspension in an adult mouse to induce neurodegeneration. An anesthetized mouse is secured in a stereotaxic frame, and a hole is drilled into the exposed skull for injection. A suspension of the Zika virus is injected into the brain tissue. The viral infection causes inflammation, leading to neurodegeneration.

Drosophila is widely used as a model system to study neurodegeneration. This protocol describes a method by which degeneration, as determined by vacuole formation in the brain, can be quantified. It also minimizes effects due to the experimental procedure by processing and sectioning control and experimental flies as one sample.

The anesthetized mouse is dissected to expose T9-T10 vertebrae, and their spinous processes are removed. The exposed spinal cord is then transected to inflict the injury.
