Cd63 Surface Expression

Cd63 surface expression is the appearance of the tetraspanin CD63 on the plasma membrane, where it serves as a measurable indicator of endosomal and lysosomal membrane trafficking. In response to cellular activation or vesicle fusion, CD63-containing late endosomal or lysosomal compartments move to and merge with the plasma membrane, increasing the amount of CD63 exposed at the cell surface. In pharmacology, measuring surface CD63 with flow cytometry, antibody-based assays, or imaging can help evaluate drug-induced changes in vesicle mobilization, secretion, and cellular activation. This readout also supports studies of extracellular vesicle biology and pharmacodynamic responses.

Here we demonstrate a protocol to carry out live cell staining that can be used to detect odorant receptors on the surface of HEK293T cells conveniently. In addition, it may also be used to assay for surface expression of other chemosensory receptors or GPCRs.
