Fluorescent Lighting

Fluorescent lighting is an electric lighting technology that produces visible light through a gas discharge rather than directly heating a filament, making it an important application of atomic and electromagnetic physics. When an electric current passes through low-pressure mercury vapor, excited mercury atoms emit ultraviolet radiation; a phosphor coating on the inside of the glass tube absorbs this radiation and converts it into visible light. A ballast regulates the current and helps initiate the discharge, while the phosphor composition influences the lamp’s color. Fluorescent lamps have been used in homes, schools, offices, and laboratories because they provide efficient, diffuse illumination, although they require careful handling because they contain mercury.

2016

Light sheet fluorescence microscopy is an excellent tool for imaging embryonic development. It allows recording of long time-lapse movies of live embryos in near physiological conditions. We demonstrate its application for imaging zebrafish eye development across wide spatio-temporal scales and present a pipeline for fusion and deconvolution of multiview datasets.

Source: Liau, E. S. et al. Visualization of Motor Axon Navigation and Quantification of Axon Arborization In Mouse Embryos Using Light Sheet Fluorescence Microscopy. J. Vis. Exp. (2018). This video demonstrates the use of light sheet fluorescence microscopy to image motor neuron projections and axon branching in transgenic mouse embryos. It outlines the steps involved in sample preparation, imaging, and 3D reconstruction of axonal branching for assessing motor neuron growth and connectivity.
