La réplication de l'ADN se produit en synthétisant de nouveaux brins d'ADN utilisant des brins existants comme modèle. Les deux nouvelles doubles hélices contiennent chacune un brin gabarit original et un brin fille nouvellement synthétisé, voila pourquoi ce processus est connu comme réplication semi-conservatrice. Pour commencer la réplication, une enzyme, l'hélicase, déroule l'hélice d'ADN et rompt les liaisons hydrogène entre les deux brins.Il sépare ensuite les brins individuels formant une structure en forme de Y, connue sous le nom de fourche de réplication, où les brins modèles sont accessibles par des enzymes supplémentaires. Une autre enzyme, la primase, ajoute de courts fragments d'ARN appelées amorces sur chaque brin modèle. Ces amorces sont essentielles pour la synthèse de l'ADN car l'ADN polymérase ne peut ajouter que des nucléotides à un brin existant.L'ADN polymérase s'ajoute aux brins filles en croissance sur les deux brins d'ADN matrice. L'ajout de nucléotides est guidé par la séquence du brin d'ADN d'origine selon les règles d'appariement ADN. La synthèse de l'un des brins filles, le brin principal, se produit dans la direction du mouvement de la fourche de réplication.L'autre brin, le brin retardé, est synthétisé dans la direction opposée. Cela conduit à la synthèse du brin principal en tant que polymère continu, alors que le brin de retard est synthétisé sous forme de fragments courts. Cela se produit parce que l'ADN ne peut être synthétisé que dans la direction 5 à 3 Avant leur ajout au polymère en croissance, les nucléotides existent sous forme de désoxyribonucléoside triphosphates, avec trois phosphates attaché au cinquième carbone sur le sucre.Ce nucléotide triphosphate libre réagit avec les groupes hydroxyle 3 C'est l'OH qui est attaché au troisième carbone du sucre à la fin du brin croissant. La réaction provoque la libération de pyrophosphate et la formation d'une liaison phosphodiester entre les deux nucléotides. Après la synthèse des nouveaux brins, la RNase H ou des variantes supplémentaires d'ADN polymérase, retirent les amorces et synthétisent l'ADN à leur place.Les espaces entre les fragments sont ensuite scellés par l'ADN ligase pour générer un brin continu. L'ajout de nucléotides se poursuit jusqu'à ce que deux fourches de réplication se rencontrent, ce qui entraîne la réplication terminée.

DNA Replication

La réplication de l'ADN implique la séparation des deux brins de la double hélice, chaque brin servant de modèle à partir duquel le nouveau brin complémentaire est copié.  Après la réplication, chaque ADN double brin comprend un brin parent ou “ ancien ” et un brin “ nouveau ”. Il s'agit de la réplication semi-conservatrice. Les molécules d'ADN qui en résultent ont la même séquence et sont divisées également en deux cellules filles. 
 Réplication dans les procaryotes 
 La réplication de l'ADN utilise un grand nombre de protéines et d'enzymes. L'enzyme hélicase sépare les deux brins d'ADN. Lorsque l'hélicase se déplace le long de l'ADN, elle sépare les deux brins pour former une structure en y, connue sous le nom de fourche de réplication. Par la suite, l'enzyme primase ajoute de courtes portions d'ARN appelées amorces pour initier la synthèse de l'ADN par l'ADN polymérase, l'enzyme responsable de la synthèse de l'ADN. Chez les bactéries, trois types principaux de polymérases d'ADN sont connus : l'ADN pol I, l'ADN pol II et l'ADN pol III. L'ADN pol III est l'enzyme requise pour la synthèse de l'ADN ; l'ADN pol I et l'ADN pol II sont nécessaires principalement pour la réparation. L’ADN pol III ajoute des désoxyribonucléotides, chacun étant complémentaire à un nucléotide sur le brin modèle, un par un au groupe 3’-OH de la chaîne d’ADN croissante. L'ADN polymérase III ne peut s'étendre que dans le sens 5’ à 3’. L'addition de ces nucléotides nécessite de l'énergie. Cette énergie est présente dans les liaisons de trois groupes phosphate fixés sur chaque nucléotide, de la même façon que l'énergie est stockée dans les liaisons phosphate de l'adénosine triphosphate (ATP). Lorsque la liaison entre les phosphates est rompue et que le diphosphate est libéré, l'énergie libérée permet la formation d'une liaison phosphodiester covalente par synthèse de déshydratation. 
 La double hélice de l’ADN est antiparallèle, c’est-à-dire qu’un brin est orienté dans le sens 5’ à 3’ et l’autre dans le sens 3’ à 5’. Au cours de la réplication, un brin, qui est complémentaire au brin d'ADN parental de 3’ à 5’, est synthétisé en continu vers la fourche de réplication parce que la polymérase peut ajouter des nucléotides dans ce sens. Ce brin synthétisé en continu est appelé le brin principal. L’autre brin, complémentaire à l’ADN parental de 5’ à 3’, s’éloigne de la fourche de réplication, de sorte que la polymérase doit revenir vers la fourche de réplication pour commencer à ajouter des bases à une nouvelle amorce, à nouveau dans le sens opposé à la fourche de réplication. Elle le fait jusqu'à ce qu'elle heurte le brin précédemment synthétisé, puis elle revient de nouveau en arrière. Ces étapes produisent de petits fragments de séquence d'ADN appelés fragments d'Okazaki, chacun séparé par une amorce d'ARN. Le brin avec les fragments d'Okazaki est connu comme le brin de retard, et sa synthèse est dite discontinue. 
 Après la synthèse par l'ADN polymérase III, les amorces sont éliminées par l'activité exonucléase de l'ADN polymérase I, et les écarts sont comblés. Les entailles qui restent entre l'ADN nouvellement synthétisé (qui a remplacé l'amorce d'ARN) et l’ADN synthétisé précédemment sont scellées par l’enzyme ADN ligase qui catalyse la formation d’une liaison covalente phosphodiester entre l’extrémité 3’-OH d’un fragment d’ADN et l’extrémité 5’ phosphate de l’autre fragment, stabilisant ainsi le squelette sucre-phosphate de la molécule d’ADN. 
 Réplication dans les eucaryotes 
 Les génomes d'eucaryotes sont beaucoup plus complexes et plus grands que les génomes de procaryotes et sont généralement composés de plusieurs chromosomes linéaires. Le génome humain, par exemple, compte 3 milliards de paires de bases par ensemble haploïde de chromosomes et 6 milliards de paires de bases sont insérées pendant la réplication. Il existe de multiples origines de réplication sur chaque chromosome d'un eucaryote ; le génome humain a 30 000 à 50 000 origines de réplication. Le taux de réplication est d'environ 100 nucléotides par seconde, soit 10 fois plus lent que la réplication des procaryotes. 
 Les étapes essentielles de la réplication chez les eucaryotes sont les mêmes que chez les procaryotes. Avant que la réplication ne puisse commencer, l'ADN doit être mis à disposition sous forme de modèle. L'ADN d'un eucaryote est fortement sur-spiralé et tassé, ce qui est facilité par de nombreuses protéines, y compris les histones. Après l'initiation de la réplication, dans un processus semblable à celui observé chez les procaryotes, l'élongation est facilitée par les ADN polymérases d'eucaryotes. Le brin principal est synthétisé en continu par l'enzyme d'eucaryotes polymérase pol δ, tandis que le brin en retard est synthétisé par pol ε. L'enzyme ribonucléase H (RNase H), au lieu d'une ADN polymérase comme dans les bactéries, supprime l'amorce d'ARN, qui est ensuite remplacée par des nucléotides d'ADN. Les espaces restant sont scellés par l'ADN ligase. 
 Comme chez les procaryotes, l'ADN polymérase des eucaryotes ne peut ajouter des nucléotides que dans le sens 5’ à 3’. Dans le brin principal, la synthèse se poursuit jusqu'à ce qu'elle atteigne l'extrémité du chromosome ou une autre fourche de réplication progressant dans la direction opposée. Sur le brin en retard, l'ADN est synthétisé en courtes portions, chacune étant initiée par une amorce séparée. Lorsque la fourche de réplication atteint l'extrémité du chromosome linéaire, il n'y a pas lieu de faire une amorce pour le fragment d'ADN à copier à l'extrémité du chromosome. Ces extrémités restent donc non appariées et, avec le temps, elles peuvent devenir progressivement plus courtes à mesure que les cellules continuent à se diviser.

DNA replication copies genetic material before a cell divides. Each original DNA strand serves as a template for a new complementary strand. The result is semiconservative replication, which means each new DNA molecule contains one old strand and one new strand. The two daughter cells then receive DNA with the same sequence.

Replication starts when helicase separates the two strands of the double helix. This creates a Y-shaped replication fork. Primase then adds short RNA primers that give DNA polymerase a place to begin DNA synthesis. In bacteria, the main DNA polymerases are DNA pol I, DNA pol II, and DNA pol III. DNA pol III carries out DNA synthesis, while DNA pol I and DNA pol II mainly help with repair.

DNA pol III adds deoxyribonucleotides one at a time to the 3'-OH end of the growing strand. It can extend DNA only in the 5' to 3' direction. The energy for this process comes from the three phosphate groups on each nucleotide. When a phosphate bond is broken and diphosphate is released, that energy helps form the phosphodiester bond in the DNA backbone.

Because the DNA double helix is antiparallel, the two template strands face opposite directions. The strand copied toward the replication fork is made continuously and is called the leading strand. The other strand is copied away from the fork. It must start over with new primers as the fork opens, so it is made in short stretches called Okazaki fragments. This strand is the lagging strand, and its synthesis is discontinuous.

After DNA pol III finishes, DNA pol I removes the RNA primers in bacteria, and the gaps are filled in. DNA ligase then seals the remaining nicks by forming covalent phosphodiester bonds between adjacent DNA fragments. This step strengthens the sugar-phosphate backbone of the new DNA molecule.

Eukaryotic DNA replication follows the same basic steps, but the genome is larger and more complex. Eukaryotic chromosomes are linear and have many origins of replication. In humans, the genome has 3 billion base pairs per haploid set, and about 6 billion base pairs are copied during replication. Human chromosomes also have about 30,000 to 50,000 origins of replication, and replication occurs at about 100 nucleotides per second, which is much slower than in prokaryotes.

Before replication begins, eukaryotic DNA must be made available as a template. It is tightly packed and supercoiled, and histones help organize this packaging. After initiation, eukaryotic DNA polymerases carry out elongation. Polymerase delta (pol δ) synthesizes the leading strand, and polymerase epsilon (pol ε) synthesizes the lagging strand. RNase H removes the RNA primers, DNA nucleotides replace them, and DNA ligase seals the final gaps. Because eukaryotic DNA polymerase also works only in the 5' to 3' direction, the lagging strand is still made in short pieces. Near the ends of linear chromosomes, there is no place to add a final primer, so these ends remain unpaired and can become shorter over time as cells keep dividing.
