Enzymes and Activation Energy

Dans une réaction chimique, l’énergie d’activation, ou Ea, est la différence d’énergie libre entre les substrats dans leur état fondamental et dans leur état de transition à haute énergie, une structure transitoire instable.
Ces réactifs doivent avoir une énergie cinétique suffisante pour entrer en collision fréquemment et subir des modifications chimiques afin de former des produits.
En raison de l’énergie d’activation d’une réaction, l’état de transition a une énergie plus élevée que les réactifs ou les produits. Il peut rapidement perdre cette énergie et retourner aux réactifs ou se transformer en produits.
Lorsque les substrats se lient à leurs enzymes, l’énergie d’activation diminue, ce qui permet à un plus grand nombre de réactifs d’atteindre l’état de transition. Cela permet de transformer davantage de molécules en produits et d’augmenter la vitesse d’une réaction catalysée par une enzyme par rapport à une réaction non catalysée.
Les enzymes accélèrent les vitesses de réaction de plusieurs façons, notamment en stabilisant l’état de transition en positionnant les substrats dans la bonne orientation ou en fournissant des environnements chimiques appropriés, tels que la charge ou le pH.

L'énergie d'activation (ou énergie libre d'activation), Ea en abrégée, est la petite quantité d'énergie nécessaire à toutes les réactions chimiques pour qu’elles se produisent. Lors des réactions chimiques, certaines liaisons chimiques se rompent et de nouvelles se forment. Par exemple, lorsqu'une molécule de glucose se décompose, les liaisons entre les atomes de carbone de la molécule se brisent. Puisqu’il s’agit de liaisons stockant de l’énergie, elles la libèrent suite à cette rupture. Cependant, la molécule doit être quelque peu déformée pour entrer dans un état permettant que ces liaisons se rompent. Un faible apport en énergie est nécessaire pour atteindre cet état instable, et hautement énergétique appelé état de transition. Pour cette raison, les molécules réactives ne restent pas longtemps dans leur état de transition mais passent très rapidement aux étapes suivantes de la réaction chimique. L’état de transition de la réaction demeure à niveau d’énergie plus élevé que celui des réactifs, et donc l’ Ea est toujours positive.

Lorsqu’une enzyme se lie à son substrat, elle forme un complexe enzyme-substrat. Ce dernier réduit l’énergie d’activation de la réaction et favorise sa progression rapide de plusieurs manières. Le complexe enzyme-substrat peut réduire l'énergie d'activation en contorsionnant les molécules du substrat de manière à faciliter la rupture des liaisons, aidant ainsi à atteindre l'état de transition. Les enzymes peuvent également réduire les énergies d’activation en participant à la réaction chimique. Les résidus d'acides aminés peuvent fournir des ions ou des groupes chimiques qui forment des liaisons covalentes avec des molécules de substrat, étape nécessaire à la réaction. Il est important de garder à l’esprit que l’enzyme reviendra toujours à son état d’origine une fois la réaction terminée. L’une des propriétés distinctives des enzymes est qu’elles restent finalement inchangées par les réactions qu’elles catalysent. Après qu’une enzyme ait catalysé une réaction, elle libère son ou ses produits.

how enzymes lower reaction energy

Enzymes lower the energy needed for a chemical reaction to begin. That starting energy is called activation energy, or free energy of activation, and it is often written as Ea. During a reaction, bonds break and new bonds form. In glucose breakdown, for example, bonds between carbon atoms break and release energy because they are energy-storing bonds.

Before a reaction can move forward, reactant molecules must reach a high-energy, unstable shape called the transition state. To get there, the molecules may need to become somewhat contorted. This small energy input is required because the transition state sits at a higher energy level than the reactants. For that reason, activation energy is always positive, and reactants do not stay in the transition state for long.

Enzymes work by binding their substrates and forming an enzyme-substrate complex. This complex helps lower activation energy and speeds up the reaction. In some cases, the enzyme shapes the substrate so bond-breaking becomes easier and the transition state is reached more quickly. In other cases, amino acid residues in the enzyme take part in the reaction by providing specific ions or chemical groups that form covalent bonds with the substrate.

After the reaction finishes, the enzyme releases its product or products. The enzyme returns to its original state and is not permanently changed by the reaction it catalyzes. That unchanged recycling is one of the key features that makes enzymes effective catalysts.
