L’adénosine triphosphate ou ATP est la monnaie d’énergie la plus importante qui alimente plusieurs processus biochimiques à l’intérieur d’une cellule vivante.
L’ATP est un composé organique constitué d’une molécule d’adénosine représentée par A, liée à trois groupes phosphate représentés par la lettre P. Les trois phosphates sont reliés les uns aux autres par deux liaisons phosphoanhydride à haute énergie. L’hydrolyse de ces liaisons peut produire environ 46 à 54 kilojoules par mole d’énergie libre, selon les conditions intracellulaires.
Étant donné que la fixation d’un groupe phosphate à une molécule d’ADP est énergétiquement défavorable, les cellules puisent de l’énergie dans la photosynthèse ou la respiration cellulaire pour former la liaison phosphoanhydride entre l’ADP et le troisième groupe phosphate.

Un exemple de couplage énergétique utilisant l’ATP implique une pompe ionique transmembranaire extrêmement importante pour la fonction cellulaire. Cette pompe sodium-potassium (pompe Na^+/K^+) chasse le sodium hors de la cellule et le potassium dans la cellule. Un pourcentage important de l'ATP d'une cellule est utilisé pour alimenter cette pompe, car les processus cellulaires importent régulièrement de grandes quantités de sodium dans la cellule et exportent de grandes quantités de potassium hors de la cellule. La pompe travaille constamment à stabiliser les concentrations cellulaires de sodium et de potassium. Pour que la pompe effectue un cycle (exportant trois ions Na^+ et important deux ions K^+), une molécule d’ATP doit être hydrolysée. Lorsque l’ATP est hydrolysé, son gamma phosphate est transféré sur la protéine de pompe.

Ce processus de liaison d’un groupe phosphate à une molécule est appelé phosphorylation. Comme dans la plupart des cas d’hydrolyse de l’ATP, un phosphate de l’ATP est transféré sur une autre molécule. Dans un état phosphorylé, la pompe Na^+/K^+ a plus d’énergie libre et est déclenchée pour subir un changement de conformation. Ce changement lui permet de libérer Na^+ vers l'extérieur de la cellule. Il lie ensuite le K^+ extracellulaire, ce qui, par un autre changement de conformation, provoque le détachement du phosphate de la pompe. Cette libération de phosphate déclenche la libération du K^+ à l’intérieur de la cellule. Essentiellement, l’énergie libérée par l’hydrolyse de l’ATP est couplée à l’énergie nécessaire pour alimenter la pompe et transporter les ions Na^+ et K^+. L'ATP effectue un travail cellulaire en utilisant cette forme fondamentale de couplage énergétique par phosphorylation.

Souvent, au cours de réactions métaboliques cellulaires, telles que la synthèse et la dégradation des nutriments, certaines molécules doivent légèrement modifier leur conformation pour devenir des substrats pour l'étape suivante de la série de réactions. Un exemple est la glycolyse, les toutes premières étapes de la respiration cellulaire. Dans cette première étape, l’ATP est nécessaire pour phosphoryler le glucose, créant ainsi un intermédiaire à haute énergie mais instable. Cette réaction de phosphorylation entraîne un changement de conformation qui permet à la molécule de glucose phosphorylée de se convertir en sucre fructose phosphorylé. Ici, la réaction exergonique de l'hydrolyse de l'ATP, couplée à la réaction endergonique de phosphorylation du glucose, constitue une étape intermédiaire dans la voie. Une fois de plus, l’énergie libérée par la rupture d’une liaison phosphate au sein de l’ATP a été utilisée pour phosphoryler une autre molécule, créant ainsi un intermédiaire instable et alimentant un changement conformationnel important.

ATP powers cellular transport and many reactions by transferring energy through phosphorylation. ATP is a highly unstable molecule. If it is not used quickly, it breaks down into ADP and inorganic phosphate, and the released energy is lost as heat.

Cells use ATP through energy coupling. This means the exergonic reaction of ATP hydrolysis is linked to an endergonic reaction so the reaction can move forward. One major example is the sodium-potassium pump, a transmembrane ion pump that is vital for cell function.

The sodium-potassium pump moves sodium out of the cell and potassium into the cell. Cells spend a large amount of ATP on this pump because sodium and potassium are regularly moved in opposite directions across the membrane. To complete one cycle, the pump exports three sodium ions and imports two potassium ions, and one ATP molecule must be hydrolyzed.

During ATP hydrolysis, the gamma phosphate is transferred to the pump protein. This is called phosphorylation, which means a phosphate group binds to a molecule. In the phosphorylated state, the pump has more free energy and changes shape. That shape change lets it release sodium outside the cell. The pump then binds potassium outside the cell, and another shape change removes the phosphate and releases potassium inside the cell.

ATP also helps start steps in cellular metabolism. In glycolysis, which is the first stage of cellular respiration, ATP phosphorylates glucose. This creates a high-energy but unstable intermediate. The phosphorylation also drives a conformational change that helps the molecule become phosphorylated fructose, which is needed for glycolysis to continue. In both examples, ATP hydrolysis is coupled to another reaction by transferring a phosphate group and powering an important change in shape.
