Le HNO2 est un acide plus faible que le HNO3. Il s’ionise pour donner des ions hydronium et nitrite et a une valeur pKde 3,37.
Comme le HNO2 est instable, il est préparé in situ en faisant réagir le NaNO2 avec un acide minéral fort, comme l’acide chlorhydrique ou l’acide sulfurique.
Dans les mêmes conditions acides, le groupe -OH de l’acide nitreux subit une protonation pour donner un ion oxonium, qui perd une molécule d’eau pour générer le cation nitrosyle ou l’ion nitrosonium.
L’ion nitrosonium électrophile peut alors réagir avec l’amine azote nucléophile.
La réaction de HNO2 avec les amines aliphatiques et aromatiques primaires, secondaires et tertiaires donne lieu à des produits différents.
Par exemple, les amines primaires, lorsqu’elles sont traitées avec HNO2, produisent des sels de diazonium, tandis que les amines secondaires produisent des N-nitrosamines.
Lorsqu’elles réagissent avec HNO2, les amines aliphatiques tertiaires subissent une protonation pour produire des sels solubles dans l’eau et des composés N-nitrosoammonium. En revanche, les arylamines tertiaires subissent une substitution aromatique électrophile pour former des composés aromatiques C-nitroso.

nitrous acid reactions with amines

Nitrous acid reacts with amines in acidic conditions, and the products depend on the type of amine. It is a key nitrogen-containing acid, but it is weaker than nitric acid. Nitrous acid has a pKa of 3.37, so it ionizes in water to form nitrite ion and hydronium ion.

Nitrous acid is unstable, so it is made in situ, or formed in the reaction mixture as needed. A sodium nitrite solution is combined with a cold aqueous acid such as hydrochloric acid or sulfuric acid. In acid, the –OH group of nitrous acid is protonated first. This gives an oxonium ion, which then loses water to form nitrosonium ion.

Nitrosonium ion is a highly reactive electrophile, meaning it seeks electrons. It reacts with the nucleophilic nitrogen atom of an amine. Primary amines form diazonium salts in this reaction, and this process is called diazotization. Secondary amines give N-nitrosamines instead.

The outcome also changes with tertiary amines. Tertiary aliphatic amines form water-soluble salts and N-nitrosoammonium compounds. Tertiary arylamines react by electrophilic aromatic substitution to make aromatic C-nitroso compounds.
