Les amines secondaires, y compris les alkyles et les aryles, lorsqu’elles sont traitées avec du NaNO2 et un acide fort, donnent des N-nitrosamines secondaires.
Rappelons qu’une solution acidifiée de NaNO2 génère du HNO2. L’instable HNO2 subit ensuite une protonation et une déshydratation ultérieure pour former l’ion nitrosonium, un électrophile.
Le cation nitrosyle, généré in situ, est attaqué par les amines secondaires nucléophiles pour former un ion N-nitrosammonium.
L’ion, lors de la déprotonation par des molécules de solvant, entraîne la formation de N-nitrosamines secondaires.
Contrairement aux N-nitrosamines primaires, dérivées d’amines primaires, les N-nitrosamines secondaires sont stables car l’absence du proton N-H empêche la poursuite de la tautomérisme et la formation éventuelle de diazonium.
Ils se séparent généralement sous forme de liquides huileux du mélange réactionnel.
En général, les N-nitrosamines secondaires ont des applications synthétiques rares. Cependant, beaucoup d’entre eux sont des cancérogènes connus que l’on trouve dans les aliments transformés ou le tabac.

Habituellement, les N-nitrosamines secondaires sont moins importantes pour une utilisation commerciale. Cependant, ils sont étudiés en raison de leurs puissantes propriétés cancérigènes. En Norvège (1962), une intoxication alimentaire chez des moutons a été attribuée à de la farine de poisson traitée au nitrite. Le nitrite de sodium est utilisé pour conserver divers types d’aliments et de viandes. Dans l’estomac, ils réagissent avec l’acide gastrique pour former de l’acide nitreux. L'acide nitreux réagit ensuite avec les amines secondaires présentes dans les aliments pour donner des N-nitrosamines hautement cancérigènes. Pour cette raison, l'utilisation du nitrite de sodium a été limitée par la FDA à 50 à 125 ppm pour la conservation de la viande. Les N-nitrosamines se trouvent également dans la fumée de cigarette et de tabac.

nitrosamine formation from secondary amines

Secondary amines can form N-nitrosamines when they react with nitrous acid. Nitrous acid is weak and unstable, so it is made in the reaction mixture from sodium nitrite and a strong acid such as hydrochloric acid or sulfuric acid. The reaction is carried out in cold conditions.

In acid, nitrous acid becomes protonated. It then loses water and forms the nitrosonium ion, a reactive electrophile. The secondary amine acts as a nucleophile and attacks this ion. That step gives an N-nitrosammonium ion.

Water then removes a proton from the ion to make the secondary N-nitrosamine. These products can be separated from the mixture as oily yellow liquids. Although they are not usually important for commercial use, they are studied because many are potent carcinogens.

The transcript also links this chemistry to food safety. In 1962, a food-poisoning outbreak in sheep in Norway was traced to nitrite-treated fish meal. Sodium nitrite is used to preserve foods and meat, but in the stomach it can form nitrous acid. That acid can react with secondary amines in food to produce highly carcinogenic N-nitrosamines, so FDA limits sodium nitrite in meat preservation to 50 to 125 ppm. N-nitrosamines are also found in cigarette and tobacco smoke.
