Les sels d’arenedizonium, lorsqu’ils sont traités avec des cycles aromatiques substitués portant des groupes activateurs, subissent une réaction de couplage pour générer des composés portant un groupe –N=N–.
Il s’agit d’une réaction de substitution aromatique électrophile, dans laquelle l’ion diazonium électrophile réagit en position para du composé aromatique nucléophile, ce qui entraîne le carbocation stabilisé par résonance.
Ensuite, la déprotonation du carbocation conduit à la formation du produit couplé.
Si la position para du cycle aromatique n’est pas disponible, le couplage se produit en position ortho.
Les composés azoïques colorés sont souvent utilisés comme colorants synthétiques et indicateurs de pH. Par exemple, le « jaune coucher de soleil » est un colorant responsable de la couleur jaune de la crème glacée.
L’orange de méthyle est utilisé comme indicateur acido-basique lors de l’analyse titrimétrique. En présence d’une solution alcaline, il se transforme en une solution de couleur jaune, tandis que dans les milieux acides, lors de la protonation, la couleur passe au rouge.

La réaction de sels d'aryldiazonium (également appelés arènediazonium) faiblement électrophiles avec des composés aromatiques hautement activés conduit à la formation de produits avec une liaison -N=N-, appelée liaison azoïque. Cette réaction, présentée sur la figure 1, est connue sous le nom de couplage diazo et se produit sans perte des atomes d'azote du sel d'aryldiazonium. Les composés aromatiques hautement activés tels que les phénols ou les arylamines favorisent la réaction de couplage diazo. Le couplage se produit généralement en position para. Cependant, si la position para est substituée, le couplage se produit à la place en position ortho.

Mécaniquement, la réaction se déroule via une réaction de substitution électrophile via le don des électrons π du noyau aromatique activé à l'azote électrophile, ce qui entraîne un carbocation stabilisé par résonance.

La figure 3 représente l'étape finale, qui implique la déprotonation du carbocation pour donner le produit diazo.

Comme le montre la figure 4, le rôle de la basicité dans la réaction de couplage diazoïque est évident. Pour les phénols, cela est plus réalisable dans des conditions légèrement alcalines, où le phénol est présent sous forme d’ions phénoxes. La formation d'ions phénoxes appréciables active le cycle aromatique, améliorant ainsi le couplage entre les ions arènediazonium et les phénols dans une solution légèrement basique. Cependant, un excès d'alcalinité de la solution (pH &gt; 10) conduit à la formation d'ions diazohydroxyde ou diazotate, qui ne réagissent pas au couplage.

Contrairement aux phénols, comme le montre la figure 5, le couplage diazo des amines se produit de manière optimale dans la plage de pH acide de 5 à 7. La réaction est pilotée par la concentration de l'ion arènediazonium, qui est maximale à ce pH.

Les composés diazo-couplés présentent un système électronique π conjugué étendu en alignant les deux anneaux aromatiques en conjugaison, ce qui les rend fortement chromophores. Cela se traduit par la couleur intense des composés azoïques appelés colorants azoïques. Des exemples de colorants azoïques comprennent le jaune d'alizarine, le jaune beurre, le rouge congo, l'orange II et le rouge para.

En plus d'être un excellent colorant, les composés diazoïques ont également été identifiés comme antibiotiques lorsque Domagk G., en 1935, a utilisé avec succès le prontosil sur sa fille pour prévenir une infection streptococcique mortelle, ce qui a marqué le début de l'ère moderne des antibiotiques synthétiques. Prontosil est inactif contre les bactéries, mais dans le corps humain, il se métabolise pour donner un composé appelé sulfanilamide, qui présente des propriétés antibactériennes.

diazo coupling and azo dye formation

Diazo coupling is the reaction that forms azo dyes from aryldiazonium salts and highly activated aromatic compounds. The product contains an —N=N— azo linkage, and the nitrogen atoms from the diazonium salt remain in the product. Phenols and arylamines are common activated partners for this reaction.

The coupling usually takes place at the para position on the aromatic ring. If the para position is already occupied, the reaction shifts to the ortho position instead. The mechanism is an electrophilic substitution reaction. The activated aromatic ring donates pi electrons to the electrophilic nitrogen, forms a resonance-stabilized carbocation, and then loses a proton to give the diazo product.

Basicity affects diazo coupling in important ways. For phenols, the reaction works best under slightly alkaline conditions, where phenoxide ions form and activate the ring. If the solution becomes too basic, with pH above 10, diazohydroxide or diazotate ions form and do not couple well.

Amines behave differently. Their diazo coupling is most effective in the acidic pH range of 5 to 7, where the concentration of the arenediazonium ion is highest. The final azo compounds have an extended conjugated pi system because the two aromatic rings are aligned in conjugation. This makes them strongly chromophoric, which gives azo dyes their intense color.

Common examples of azo dyes include alizarin yellow, butter yellow, congo red, orange II, and para red. Diazo compounds are also linked to medicine. In 1935, Domagk used prontosil to help prevent a lethal streptococcal infection in his daughter, which marked the start of the modern era of synthetic antibiotics. Prontosil itself is inactive, but in the body it is converted to sulfanilamide, the antibacterial form.
