Rappelons que l’alkylation directe de l’ammoniac donne un mélange de 1°-, 2°-, et 3°--amines. Pour fabriquer exclusivement des amines primaires, on utilise du phtalimide, une forme protégée d’ammoniac composée d’un seul proton acide N-H capable de substitution d’un seul alkyle.
La synthèse de Gabriel utilise le phtalimide, une base, et l’hydrazine pour convertir les halogénures d’alkyle en amines primaires.
Dans un premier temps, le proton N-H du phtalimide est extrait par une base, ce qui donne un anion nucléophile stabilisé par résonance.
L'anion attaque l'halogénure d'alkyle de manière SN2 pour donner du phtalimide N-alkyle, qui, malgré le doublet non liant de l'azote, ne s'alkyle pas en raison de sa nucléophilie réduite.
Dans l’étape suivante, l’hydrazine attaque l’un des groupes carbonyles, effectuant une substitution acyle nucléophile pour cliver la liaison C-N.
Un transfert intramoléculaire de protons N-N, suivi d’une autre substitution d’acyle nucléophile par le -NH2 de l’hydrazine n’ayant pas réagi, forme un composé cyclique chargé tout en expulsant simultanément l’amine avec une charge négative sur l’azote.
L’azote négatif déprotone l’azote positif pour donner une amine primaire et un hydrazide phtalimide stable.

L'alkylation directe n'est pas une méthode appropriée pour synthétiser des amines car elle produit des produits polyalkylés. La synthèse Gabriel est la méthode la plus privilégiée pour fabriquer exclusivement des amines primaires. La méthode utilise du phtalimide, qui contient une forme protégée d'azote qui ne participe à l'alkylation qu'une seule fois pour donner principalement des amines primaires.

Des bases fortes comme NaOH ou KOH déprotonnent le phtalimide pour former l'anion correspondant, qui agit comme un nucléophile. De plus, l'anion attaque un halogénure d'alkyle pour donner du N-alkylphtalimide. Ce composé double amide réagit avec l'hydrazine sous l'influence de la chaleur pour produire des amines primaires. Alternativement, le N-alkylphtalimide peut également subir une hydrolyse en présence d'une base ou d'un acide pour produire des amines primaires.

Gabriel synthesis is a selective way to make primary amines. It avoids direct alkylation, which can create polyalkylated products. For that reason, it is a preferred method when the goal is to make only one alkylated amine product.

The reaction starts with phthalimide, which holds nitrogen in a protected form. A strong base such as NaOH or KOH removes a hydrogen from phthalimide and forms an anion. This anion acts as a nucleophile and attacks an alkyl halide, giving N-alkylphthalimide.

The N-alkylphthalimide can then be heated with hydrazine to release the primary amine. It can also be hydrolyzed with either a base or an acid to form the same product. Because the reaction follows an SN2 mechanism, unhindered primary and secondary alkyl halides are the best substrates. Gabriel synthesis is also linked to the preparation of amphetamine, a stimulant drug that was used during World War II as an appetite suppressant.
