Secondary Messengers in Hormone Action

Les hormones hydrosolubles ne peuvent pas se déplacer à travers la membrane plasmique, elles utilisent donc des récepteurs protéiques qui traversent la membrane pour déclencher des voies de signalisation intracellulaires.
Ces voies activent des seconds messagers à l’intérieur de la cellule, notamment l’AMPc ou les ions calcium.
De nombreuses hormones se lient aux récepteurs transmembranaires couplés aux protéines G qui se lient aux protéines G régulatrices. Les protéines G peuvent alors activer des enzymes telles que l’adénylyl cyclase ou la phospholipase C.
L’adénylyl cyclase convertit l’ATP en AMPc, ce qui peut activer les protéines kinases. Les kinases sont des enzymes qui ajoutent des groupes phosphate à d’autres protéines, initiant une cascade de phosphorylation.
L’épinéphrine, le glucagon, l’hormone parathyroïdienne et l’hormone lutéinisante agissent par l’intermédiaire de l’AMPc.
La phospholipase C décompose le phospholipide membranaire PIP2 en IP3 et DAG.
L’IP3 se déplace ensuite vers la membrane endoplasmique et se lie à un canal calcique IP3-dépendant, provoquant une libération d’ions calcium dans le cytoplasme. Le changement de concentration en calcium peut induire des effets physiologiques tels que la contraction musculaire.
L’ocytocine et les hormones régulatrices hypothalamiques amplifient les signaux par le biais de PIP2 et d’ions calcium.

Water-soluble hormones signal through cell receptors and second messengers. Because these hormones cannot cross the plasma membrane, they bind to protein receptors that span the membrane and start signaling inside the cell. The response often depends on small messenger molecules such as cAMP or calcium ions.

Many of these hormones use G protein-coupled receptors, or GPCRs. These membrane receptors connect to regulatory G proteins, which then activate enzymes such as adenylyl cyclase or phospholipase C. Adenylyl cyclase changes ATP into cAMP. cAMP activates protein kinases, which add phosphate groups to other proteins and start a phosphorylation cascade.

Several hormones use the cAMP pathway. Epinephrine, glucagon, parathyroid hormone, and luteinizing hormone are examples. Epinephrine binds the beta-adrenergic receptor and triggers the GPCR signaling pathway that helps control the fight-or-flight response.

Other hormones signal through phospholipase C and calcium. Phospholipase C breaks the membrane phospholipid PIP2 into IP3 and DAG. IP3 moves to the endoplasmic membrane and binds an IP3-gated calcium channel, which releases calcium ions into the cytoplasm. The rise in calcium can produce effects such as muscle contraction. Oxytocin and hypothalamic regulatory hormones use this PIP2 and calcium signaling route to amplify their signals.

The central nervous system is composed of the brain and spinal cord. The peripheral nervous system includes the remaining nervous tissue. A neuron consists of dendrites, a cell body and an axon. Neurons conduct electrical impulses along their membranes and at synapses. Brain cells can detect and sometimes respond to these impulses. Neuroglial cells help to support neural function. The brain consists of three major parts: the cerebrum, cerebellum and brainstem. The cerebrum is divided into lobes and hemispheres. Functions of the cerebrum that may be explored include voluntary muscle control, memory, sensory perception, emotions and speech. The cerebellum is primarily responsible for balance and coordination. The brainstem, a part of the autonomic nervous system, includes structural divisions that perform basic life functions such as breathing and heart rate. The spinal cord is a continuation of the brainstem. The spinal cord is a bundle of nerve tracts that transmits nerve signals between the brain and the body through electrical impulses. Nerves are bundles of neurons that transmit impulses between the peripheral and central nervous systems. The study of nerves can include sciatic, cranial and spinal nerves. Supporting structures of the central nervous system include the meninges and cerebrospinal fluid which protect the central nervous system. Processes of the nervous system are action potential propagation, simple nerve pathways (reflex arc) and neurotransmitter function. Homeostatic imbalances are explored. These include, but are not limited to, the effects of drugs, mental illnesses, spinal injuries, concussions, meningitis and multiple sclerosis (MS). Investigations are used to understand and explain the nervous system in a variety of inquiry and design scenarios that can incorporate evolutionary concepts, scientific reasoning, comparative analysis, communication skills and real-world applications.
