Exosome Biomarker Analysis

Exosome biomarker analysis is the study of molecular cargo carried by exosomes, small extracellular vesicles released by cells, to identify indicators of biological state or disease. The process typically involves isolating exosomes from biofluids or cell cultures, characterizing their size and surface markers, and analyzing enclosed proteins, lipids, or nucleic acids using methods such as immunoassays, mass spectrometry, or sequencing. In bioengineering, these profiles support noninvasive diagnostics, disease monitoring, therapeutic evaluation, and the design of exosome-based delivery systems. Standardized isolation and sensitive detection remain important for distinguishing biologically meaningful signals from contaminants and sample variability.

Exosomes are microvesicular structures found within biofluids that potentially carry important disease discriminatory biomarkers. Here, a novel method is used to specifically extract exosomes and rapidly test the exosomal cargo for both RNA/protein targets following the disruption of exosomes using non-uniform electric cyclic square waves.

In this video, we demonstrate gold immunostaining of biomarkers localized within the lumen region of ultrathin exosomal sections. This technique enables early diagnosis of diseases by visualization of target molecules within the exosomes secreted by the diseased cells.

These methods are adaptable for exosome isolation from both cell culture media and biological fluids, and can beyond analysis of molecular content also be useful in functional studies.
