Prothrombotic Tendency

Prothrombotic tendency is an increased propensity for inappropriate blood clot formation, a condition that matters because thrombosis can obstruct vessels or compromise blood-contacting medical devices. It arises when platelet activation and the coagulation cascade outweigh natural anticoagulant and fibrinolytic mechanisms, promoting thrombin generation, fibrin formation, and clot growth. In bioengineering, understanding this imbalance guides the design of hemocompatible biomaterials, vascular grafts, catheters, and extracorporeal systems, as well as assays that evaluate platelet adhesion and coagulation under flow. Controlling prothrombotic responses can improve device safety, support disease modeling, and inform strategies for preventing clot-related complications.

Thrombus Profiling Assay: A Microfluidics-Based Platform for Comprehensively Characterizing Biomechanical Thrombogenesis

This work describes a microfluidic assay that uses shear stress to trigger thrombus formation in blood and characterizes the thrombus by multiple dimensions of readout. The assay can measure the prothrombotic tendency of blood samples, and is thus useful in disease diagnosis, drug discovery, as well as basic mechanistic studies related to thrombosis.
