Clostridiales Order

The Clostridiales order is a diverse group of bacteria, including many anaerobic, endospore-forming species, that influences ecosystems, animal health, and biotechnology. When conditions become unfavorable, members can form resistant endospores, while under oxygen-limited conditions they commonly obtain energy through fermentation of organic compounds. Their metabolic activities support decomposition and nutrient cycling, contribute to host microbiomes, and enable industrial production of solvents and other chemicals. However, some species produce potent toxins or cause diseases such as botulism and tetanus, making Clostridiales important in microbiology, environmental biology, clinical research, and the study of microbial evolution.

Lentiviral Mediated Gene Silencing in Human Pseudoislet Prepared in Low Attachment Plates

A protocol to create gene modified human pseudoislets from dispersed human islet cells that are transduced by lentivirus carrying short hairpin RNA (shRNA) is presented. This protocol utilizes readily available enzyme and culture vessels, can be performed easily, and produces genetically modified human pseudoislets suitable for functional and morphological studies.
