E Coli Production

E. coli production is the use of Escherichia coli cells to manufacture biological products, making this bacterium an important platform in biotechnology and molecular biology. Researchers introduce recombinant DNA, often carried on a plasmid, into engineered cells; selected cultures are grown in controlled media, and gene expression can be induced to produce a target protein or other compound. After cultivation in bioreactors, the product is harvested and purified from the cells or growth medium. E. coli production supports the manufacture of research reagents, enzymes, vaccines, and therapeutic proteins while providing a practical system for studying gene expression, protein synthesis, and metabolic engineering.

Here, we describe a method to express and purify high quality norovirus protruding (P) domains in E. coli for use in X-ray crystallography studies. This method can be applied to other calicivirus P domains, as well as non-structural proteins, i.e., viral protein genome-linked (VPg), protease, and RNA dependent RNA polymerase (RdRp).
