Eutrophic Lakes

Eutrophic lakes are nutrient-rich freshwater ecosystems characterized by high concentrations of nitrogen and phosphorus, abundant biological production, and often reduced water clarity. Nutrient enrichment stimulates algal and aquatic plant growth; when this biomass dies, microbial decomposition consumes dissolved oxygen, potentially creating hypoxic or anoxic conditions that stress or eliminate fish and other aquatic organisms. In biology, eutrophic lakes serve as important systems for studying nutrient cycling, food webs, algal blooms, and ecosystem change. Their condition also guides water-quality monitoring and management strategies designed to reduce nutrient inputs, protect biodiversity, and restore aquatic habitats.

Laboratory Estimation of Net Trophic Transfer Efficiencies of PCB Congeners to Lake Trout (Salvelinus namaycush) from Its Prey

A technique for laboratory estimation of net trophic transfer efficiency of polychlorinated biphenyl (PCB) congeners to piscivorous fish from their prey is presented. To maximize applicability of the laboratory results to the field, the piscivorous fish should be fed prey fish that are typically eaten in the field.

Lake eutrophication is a water quality issue worldwide, making the need to identify and control nutrient sources critical. Laboratory determination of phosphorus release rates from sediment cores is a valuable approach for determining the role of internal phosphorus loading and guiding management decisions.

Microbial eukaryotes are both a source of photosynthetically-derived carbon and top predatory species in permanently ice-covered Antarctic lakes. This report describes an enrichment culture approach to isolate metabolically versatile microbial eukaryotes from the Antarctic lake, Lake Bonney, and assesses inorganic carbon fixation potential using a radioisotope assay for Ribulose-1,5-bisphophate carboxylase oxygenase (RubisCO) activity.

Detection of Tilapia Lake Virus Using Conventional RT-PCR and SYBR Green RT-qPCR

This protocol diagnoses Tilapia Lake Virus (TiLV) in tilapia tissues using RT-PCR methodologies. The entire method is described from tissue dissection to total RNA extraction, followed by cDNA synthesis and detection of TiLV by either conventional PCR or quantitative PCR using dsDNA binding a fluorescent binding dye.
