Hedgehog Pathway Inhibition

Hedgehog pathway inhibition is the suppression of a conserved cell-signaling system that regulates embryonic development, tissue maintenance, and cell fate, making it important for understanding abnormal growth. In the canonical pathway, blocking Hedgehog ligand binding to Patched or inhibiting Smoothened prevents downstream activation of GLI transcription factors, reducing expression of target genes that promote proliferation and differentiation. In biology and medicine, pathway inhibitors help clarify how signaling controls development and tissue repair while providing experimental tools for studying pathway dysregulation. They also support investigations of cancers and other disorders in which excessive Hedgehog activity contributes to disease.

Hedgehog Signaling Pathway

The Hedgehog gene (Hh) was first discovered due to its control of the growth of disorganized, hair-like bristles phenotype in Drosophila, much like hedgehog spines. Hh plays a crucial role in the development of organs and the maintenance of homeostasis in both invertebrates and vertebrates. However, while Drosophila has only one Hh protein, mammals have multiple functional Hedgehog proteins - Sonic (Shh), Desert (Dhh), and Indian Hedgehog (Ihh). All of these homologous proteins have adapted to...

The present protocol describes the establishment of a membranous nephropathy (MN) animal model, and how Kemeng Fang's inhibition reduces MN rat podocyte apoptosis by activating the PI3K/AKT signaling pathway.

Enzyme Inhibition

Inhibitors are molecules that reduce enzyme activity by binding to the enzyme. In a normally functioning cell, enzymes are regulated by a variety of inhibitors. Drugs and other toxins can also inhibit enzymes. Some inhibitors bind to the enzyme’s active site, while others inhibit enzymatic activity by binding to other sites on the protein structure. Competitive inhibitors occupy the active site of enzymes, making them unable to accommodate the substrate. However, sufficiently high...
