Liver Tissue Ablation

Liver tissue ablation is a minimally invasive method that destroys targeted liver tissue while limiting injury to surrounding structures, making it important for studying and treating localized disease. Depending on the approach, probes deliver thermal energy, extreme cold, or chemical agents that disrupt cell membranes, denature proteins, or produce coagulative necrosis within the treatment zone. In cancer research, liver tissue ablation supports investigations of tumor destruction, local recurrence, tissue repair, and immune responses after treatment. It also provides a treatment option for selected patients with primary or metastatic liver tumors, particularly when surgery is unsuitable or preservation of liver function is essential.

To help assess the molecular mechanisms underlying zebrafish biliary-driven liver regeneration, we established a liver injury model in which the nitroreductase-expressing hepatocytes are genetically ablated upon metronidazole treatment. In this protocol, we describe how to adeptly manipulate, monitor and analyze hepatocyte ablation and biliary-driven liver regeneration.

This article describes real-time monitoring of HIFU ablation in canine liver with high frame rate ultrasound imaging using diverging and plane wave imaging. Harmonic Motion Imaging for Focused Ultrasound is used to image the decrease of acoustic radiation force induced displacement in the ablated region.

The entire procedure is detailed, including pretreatment planning and imaging studies, anesthesia, adjuvant techniques to facilitate a percutaneous approach, imaging guidance of the ablation device to the tumor, thermal treatment, post-treatment care and follow-up.

Here, we describe a protocol for quantitatively imaging metals in thin sections of mouse neurological tissue.
