Developing Chamber

A developing chamber is a sealed vessel used to develop thin-layer chromatography (TLC) plates by separating components of a chemical mixture. Inside the chamber, a mobile-phase solvent rises through the stationary phase by capillary action; compounds migrate at different rates according to their solubility in the solvent and interactions with the adsorbent. Solvent vapor saturation helps produce consistent movement and well-defined spots. In chemistry, developing chambers support reaction monitoring, mixture identification, and method optimization, while the resulting separation provides qualitative information about sample composition and can guide subsequent purification or analytical procedures.

This protocol describes the visualization of biofilm development following exposure to host-factors using a slide chamber model. This model allows for direct visualization of biofilm development as well as analysis of biofilm parameters using computer software programs.

We present the protocols to examine mouse heart development using whole mount epifluorescent microscopy on mouse embryos dissected from ventricular specific MLC-2v-tdTomato reporter knock-in mice. This method allows us to directly visualize each stage of the ventricular formation during mouse heart development without labor-intensive histochemical methods.

Construction of Vapor Chambers Used to Expose Mice to Alcohol During the Equivalent of all Three Trimesters of Human Development

We demonstrate the construction of alcohol vapor chambers using readily available materials that simultaneously house 6 mouse cages. We further describe their use in a mouse model of fetal alcohol exposure equivalent to all 3 trimesters of human pregnancy. This paradigm exposes animals during gestation and postnatal days 1-12.

Development of a Nose-only Inhalation Toxicity Test Chamber That Provides Four Exposure Concentrations of Nano-sized Particles

A nose-only inhalation toxicity chamber capable of testing inhalation toxicity at four different exposure concentrations was designed and validated for flow field uniformity and cross-contamination between the exposure ports for each concentration. Here, we present a protocol to confirm that the designed chamber is effective for inhalation toxicity testing.
