Doubly Magic Nuclei

Doubly magic nuclei are atomic nuclei in which both the proton number and neutron number occupy complete nuclear shells, making them key benchmarks for understanding nuclear structure. In the nuclear shell model, strong spin-orbit coupling organizes nucleons into energy levels; when both types of nucleons fill a closed shell, the nucleus generally has a near-spherical shape, enhanced stability, and unusually high excitation energies. Examples such as helium-4, oxygen-16, calcium-40, calcium-48, and lead-208 help researchers test models of nuclear forces, shell evolution, and isotope stability. Their measured properties also inform studies of exotic nuclei and the limits of the nuclear landscape.

Differential Imaging of Biological Structures with Doubly-resonant Coherent Anti-stokes Raman Scattering (CARS)

A combination of three single wavelength short-pulsed lasers is used to generate coherent anti-Stokes Raman scattering (CARS) and doubly-resonant CARS (DR-CARS). The difference between these signals provides enhanced sensitivity for otherwise difficult to detect coherent Raman signals, enabling imaging of weak Raman scatterers.

This video describes the protocol to isolate single-nuclei from fresh frozen glioma tissue. The isolated single-nuclei can be used as specimens for sequencing techniques such as single-nuclei RNA (snRNA) sequencing and single-nuclei Assay for Transposase-Accessible Chromatin (snATAC) sequencing to obtain deeper insights into inter- and intra-tumor heterogeneity observed in glioma of the central nervous tissues.
