Kidney Capsule Grafting

Kidney capsule grafting is a transplantation technique in which tissue or an organ rudiment is placed beneath the capsule surrounding a kidney, creating a supportive site for growth and analysis. The graft is held against the kidney surface, where the host environment can support its survival, vascularization, and continued development while preserving tissue interactions. In developmental biology, this method enables researchers to examine embryonic organ formation, signaling between tissues, and the effects of genetic or experimental manipulation in vivo. It also supports studies of regenerative potential and tissue differentiation by providing a controlled setting for monitoring graft growth and organization.

Our protocol was developed to cleanly and easily deliver islets or cells under the kidney capsule of mice. Cells are concentrated into pellets in the final tubing used for transplanting the cells under the kidney capsule. The ease of this technique reduces stress to the cells and the mouse.

The severe organ shortage has resulted in increased use of marginal kidney grafts for transplantation. This has triggered interest in alternative storage methods, since marginal grafts especially tolerate cold storage poorly. The technique of normothermic ex vivo kidney perfusion (NEVKP) represents a novel preservation method for kidney grafts prior to transplantation.
