Micro-aspirator Assembly

Micro-aspirator assembly is the preparation of a small-volume aspiration system used to collect, transfer, or position cells and biological material with controlled suction. The assembly connects a pressure source, flexible tubing, a pipette holder, and a fine glass capillary or micropipette so that changes in pressure draw material into the tip without requiring direct contact. In genetics, this setup supports precise manipulation of embryos, individual cells, nuclei, or other samples during micromanipulation procedures. Reliable assembly and leak-free connections improve control, reduce sample loss, and help researchers perform reproducible genetic analysis, cell isolation, and experimental transfers.

Here, we describe a single cell micro-aspiration method for the separation of infected amoebae. In order to separate viral subpopulations in Vermamoeba vermiformis infected by Faustoviruses and unknown giant viruses, we developed the protocol detailed below and demonstrated its ability to separate two low-abundance novel giant viruses.

In this protocol we demonstrate how to fabricate and condition tetrodes for use with a micro-drive array, which was designed for chronic electrophysiological recordings in rats.
