Avicel Overlay

Avicel overlay is a semisolid microcrystalline-cellulose medium used in virology to confine viral spread across cultured cell monolayers, enabling plaques to form and be quantified. After inoculation, Avicel particles create a porous, viscous layer that limits movement of newly produced virions while permitting nutrients and gases to reach the cells; repeated infection therefore remains localized. Researchers use the resulting plaques to measure infectious virus, compare replication among strains or conditions, and evaluate antiviral or neutralizing-antibody activity. In immunology and infection research, this assay links visible cell damage with quantitative measurements of viral infectivity.

Plaque assays are the gold standard for viral quantification, utilizing entrapping overlays on host cellular monolayers to determine viral titers. While various semisolid overlays have traditionally been used, here we demonstrate plaque techniques comparing semisolid overlays to a novel liquid microcrystalline cellulose among several families of viruses.

Testing protein-protein interaction is indispensable for dissection of protein functionality. Here, we introduce an in vitro protein-protein binding assay to probe a membrane-immobilized protein with a soluble protein. This assay provides a reliable method to test interaction between an insoluble protein and a protein in solution.
