Humanized Blt Mouse

A humanized BLT mouse is an immunodeficient mouse model reconstituted with human bone marrow, liver, and thymus tissues to support development of a functional human immune system. In this model, human hematopoietic stem cells engraft and generate immune cells, while the implanted human thymic tissue promotes human T-cell maturation and education. Humanized BLT mice enable researchers to study human-specific immune responses, host-pathogen interactions, and infections such as HIV in vivo. They also provide a platform for evaluating vaccines, antiviral therapies, and other immunological interventions when conventional mouse models cannot accurately reproduce human biology.

The generation and characterization of tumor specific T cells using humanized mice is described here. Human thymic tissue and genetically modified human hematopoietic stem cells are transplanted into immunocompromised mice. This results in the reconstitution of an engineered human immune system allowing for in vivo examination of anti-tumor immune responses.
