Metabolic Inhibitors

Metabolic inhibitors are compounds that suppress cellular metabolism by blocking enzymes, transporters, or other components needed to generate energy and biosynthetic precursors. Depending on the inhibitor, it may bind an enzyme’s active site, compete with a substrate, or disrupt nutrient uptake, thereby changing metabolite levels and restricting pathway activity. In immunology and infection research, these agents help distinguish metabolic requirements of pathogens from those of immune cells, including the shifts that support activation, proliferation, and antimicrobial function. They are used to investigate host–pathogen interactions, validate therapeutic targets, and assess how metabolic disruption influences infection outcomes and immune responses.

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

Eukaryotic Transcription Inhibitors

2020

Certain biochemical processes, such as embryonic development and cell growth regulation, depend on the repression of specific genes. DNA binding proteins known as eukaryotic transcription inhibitors regulate the repression of gene expression in eukaryotes. The presence of these inhibitors at the required location and time in the cell is triggered by the presence of hormones and additional signals from other cells. Eukaryotic transcription inhibitors usually contain two distinct domains, a DNA...

Characterization of Metabolic Status in Nonhuman Primates with the Intravenous Glucose Tolerance Test

The goal of this protocol is to present a standard method to perform intravenous glucose tolerance tests (IVGTTs) to assess glycemic control in nonhuman primates and assess their metabolic status from healthy to dysmetabolic.

Assessing Cellular Target Engagement by SHP2 (PTPN11) Phosphatase Inhibitors

The ability to assess target engagement by candidate inhibitors in intact cells is crucial for drug discovery. This protocol describes a 384 well format cellular thermal shift assay that reliably detects cellular target engagement of inhibitors targeting either wild-type SHP2 or its oncogenic variants.

An Introduction to Cell Metabolism

2023

In cells, critical molecules are either built by joining together individual units like amino acids or nucleotides, or broken down into smaller components. Respectively, the reactions responsible for this are referred to as anabolic and catabolic. These reactions require or produce energy typically in the form of a “high-energy” molecule called ATP. Together, these processes make up “Cell Metabolism,” and are hallmarks of healthy, living cells.JoVE’s introduction to cell metabolism briefly...
