Nlrp3 Activation

NLRP3 activation is the cellular process that assembles the NLRP3 inflammasome, an innate immune signaling platform that detects infection and tissue stress. Diverse danger signals, including potassium efflux, mitochondrial dysfunction, and lysosomal damage, promote NLRP3 oligomerization with the adaptor ASC, activating caspase-1. Caspase-1 then processes pro-interleukin-1β and pro-interleukin-18 into inflammatory cytokines and can trigger pyroptosis, an inflammatory form of cell death. In immunology and infection research, studying this pathway clarifies host defense mechanisms and inflammatory pathology, while informing investigations of vaccines, infectious diseases, and therapies for disorders driven by excessive inflammasome activity.

Dendritic cells (DCs) secrete IL-1β in response to TLR8 recognition of synthetic purine, R848, followed by NLRP3 inflammasome activation with nigericin, therefore, IL-1β can be used to measure NLRP3 inflammasome activity. Intracellular cytokine staining, immunoblotting, and ELISA are used to accurately measure NLRP3 inflammasome priming and activation via IL-1β expression.

Detection of Inflammasome Activation via Fluorescence Microscopy

This video demonstrates a method to detect NLRP3 inflammasome activation in macrophages using fluorescent reporter probes. The cells are treated with potassium ionophores and glycine to induce inflammasome assembly and caspase-1 activation. Fluorescent reporter probes visualize activated caspase-1, and cells are stained with a fluorescent dye for nuclei visualization.

This report describes a method for measuring Drosophila larval activity using the TriKinetics Drosophila Activity Monitor. The device employs infrared beams to detect movements of up to 16 individual animals. Data can be analyzed to represent motion parameters including rates and the positions of the animals within the assay chambers.

Biological Catalysts All living organisms continuously perform numerous biochemical reactions to sustain their presence. Most of these reactions require an input of energy to start, which is called the activation energy. Catalysts are chemicals that lower the activation energy. Even though catalysts facilitate a chemical reaction, they are not consumed by it. This means a catalyst can facilitate a specific chemical reaction over and over in succession, increasing the rate of that reaction.
