Viral Titer Production

Viral titer production is the process of generating virus stocks and determining their concentration, typically as infectious units per volume, for controlled laboratory use. Researchers propagate a virus in susceptible host cells, collect and clarify the resulting material, and quantify infectivity with assays such as plaque formation or the tissue culture infectious dose method. In immunology and infection research, standardized viral titers support reproducible infection experiments, host-pathogen studies, antiviral testing, and vaccine evaluation. Accurate titration also helps researchers compare experimental conditions, control the multiplicity of infection, and interpret immune or cellular responses with greater confidence.

Recombinant adeno-associated virus (rAAVs) vectors are becoming increasingly valuable for in vivo studies in animals. We describe how rAAVs can be produced in the laboratory and how these vectors can be titered to give an accurate reading of the number of infectious particles produced.

Exp., (2022)This video demonstrates a crystal violet staining method to visualize cytopathic effects in virus-infected cell cultures. The pattern of stained and unstained wells across serial dilutions enables rapid, cost-effective estimation of the viral infectious dose.

Treatment of Platelet Products with Riboflavin and UV Light: Effectiveness Against High Titer Bacterial Contamination

The storage conditions of platelet products create an ideal environment for bacterial contaminants to multiply to dangerous levels. The goal of this study was to use a colony forming assay to evaluate a riboflavin based pathogen reduction process against high titer bacterial contamination in human platelet products.

We describe the enzyme-linked lectin assay (ELLA) for measuring influenza neuraminidase (NA)-inhibition antibody titers in sera. The assay uses peanut agglutinin to quantify galactose residues that become accessible when NA removes sialic acid from fetuin-coated, 96-well plates.

Phages have since been widely recognized for their therapeutic value (3) and their influence on human (4), as well as global, ecosystems (5). Current concerns have...
