Hepatocyte Transplantation

Hepatocyte transplantation is a cell-based therapy in which functional liver cells are introduced into a patient to replace or support impaired hepatic activity. After delivery, transplanted hepatocytes must survive, engraft within host tissue, establish access to nutrients and oxygen, and perform essential metabolic, synthetic, and detoxification functions. The approach is being studied for acute or chronic liver failure and inherited metabolic disorders, where healthy cells may provide temporary support or correct specific biochemical defects. By offering a less invasive alternative to whole-organ transplantation in selected cases, hepatocyte transplantation also advances research on cell therapy, tissue regeneration, and liver disease treatment.

We have described a protocol for performing partial hepatectomy (PHx) and cell transplantation via spleen in NOD.SCID (NOD.CB17-Prkdcscid/J) mice. In this protocol, an incision is made to expose and resect the left lobe of the liver followed by another incision for the intrasplenic transplantation of cells.

A modified two-step collagenase perfusion procedure for isolation of human hepatocytes is described. This method can also be applied to other mammalian livers. The isolated hepatocytes are available in high yield and viability, making them a suitable model for scientific research in areas such as liver regeneration, pharmacokinetics and toxicology.
