Ovarian Graft Thawing

Ovarian graft thawing is the controlled rewarming of cryopreserved ovarian tissue before transplantation or laboratory evaluation, a critical step in fertility preservation and reproductive medicine. The process gradually restores tissue temperature while limiting ice-crystal damage and osmotic stress, then removes cryoprotective agents through carefully managed dilution steps to support follicle and stromal-cell viability. Proper thawing helps preserve the graft’s structural integrity and endocrine or reproductive potential, allowing assessment before implantation. This technique contributes to fertility-restoration strategies for patients whose ovarian function may be compromised by disease or treatment, while also supporting research on ovarian tissue cryopreservation and graft performance.

We describe an ovarian fad pad transplantation assay suitable for studies of normal and transformed epithelia of the female reproductive tract. The mouse fat pad allows transplantation of large tissue fragments, is easily accessible for surgery and imaging, and provides the most favorable native environment for tissues of Müllerian origin.
