Placental Explant Culture

Placental explant culture is an ex vivo technique that maintains small pieces of placental tissue under controlled laboratory conditions to study human placental function. In culture, tissue fragments retain local cell–cell and cell–matrix interactions, allowing trophoblast and other placental cells to release soluble factors and respond to defined environmental or experimental conditions. This model supports investigation of trophoblast invasion, vascular development, transport, and inflammatory signaling while preserving aspects of tissue organization that isolated-cell systems may lose. In medicine, placental explants help researchers examine disorders such as preeclampsia, evaluate molecular pathways and treatments, and generate clinically relevant evidence about maternal–fetal health.

A simple method to establish primary human placental (villous) and decidual organ cultures is described. Villous and decidual organ cultures are invaluable tools for studying pathogenesis at the human maternal-fetal interface. Infection with the facultative intracellular bacterium Listeria monocytogenes is demonstrated.
