Scratch Response Assessment

Scratch response assessment is a method for evaluating how cells or tissues respond to a controlled mechanical injury, providing insight into repair and regenerative capacity. In a typical in vitro scratch assay, researchers create a reproducible gap in a confluent cell layer and track its closure over time as cells migrate, proliferate, or reorganize at the wound edge. Measuring changes in gap area or closure rate helps characterize wound-healing behavior and identify factors that enhance or inhibit repair. In medicine, this approach supports studies of tissue regeneration, disease-related healing defects, and the screening of compounds that influence cellular recovery.

Fragility Assessment of Bovine Cortical Bone Using Scratch Tests

This study assesses the fracture toughness of bovine cortical bone at the sub-meso levels using microscopic scratch tests. This is an original, objective, rigorous, and reproducible method proposed to probe fracture toughness below the macroscopic scale. Potential applications are studying changes in bone fragility due to diseases like osteoporosis.

The ability to monitor T cell responses in detail in vivo is important for the development of our understanding of the immune response. Here we describe the use of fluorescent target arrays (FTAs) in an in vivo T cell assay that assesses >250 parameters simultaneously by flow cytometry.

We have developed a simple and reproducible protocol to access stomatal response to live bacteria. This method minimizes wounding and manipulation of the leaf as compared to the use of epidermal peels reported previously.

We present a cost-effective method to the scratch migration assay that provides a new approach for determining cell migration without the use of equipment-intensive methods. While fibroblasts were used in this protocol, it can be adapted and utilized to study additional cell types and influences on cell migration.

This protocol describes an easily adaptable migration assay that uses rigorous statistics to quantify changes in MSC migratory capacity after damage.
