Fluorescent Cargo Import

Fluorescent Cargo Import is an experimental approach for measuring how labeled molecules enter and move through cells, providing a visible readout of intracellular transport. In neuroscience, researchers attach fluorescent markers to cargo and monitor its uptake, vesicular trafficking, and distribution with fluorescence microscopy, often comparing signal intensity or localization over time. The method helps characterize endocytic pathways, receptor-mediated transport, and axonal cargo delivery in neurons. By revealing where and when cargo enters neural cells, fluorescent cargo import assays support studies of synaptic maintenance, neuronal connectivity, and transport defects associated with neurological disease.

Identification of Kinesin-1 Cargos Using Fluorescence Microscopy

Here, a protocol is presented to identify Kinesin-1 cargos. A motorless mutant of the Kinesin-1 heavy chain (KIF5B) aggregates in the cytoplasm and induces aggregation of its cargos. Both aggregates are detected under fluorescence microscopy. A similar strategy can be employed to identify cargos of other motor proteins.

Here we describe a new method to study protein import into isolated chloroplasts under stress. The method is rapid and straightforward, and can be applied to study the consequences of different stress conditions for chloroplast protein import, and the corresponding regulatory mechanisms.
