Head Mount Microscope

A head-mounted microscope is a miniaturized imaging system secured to an animal’s head to record neural activity while it moves freely, making it valuable for linking brain function with natural behavior. In neuroscience, the device typically directs excitation light through a miniature objective onto fluorescent indicators, then collects emitted signals with a camera or fiber-free sensor; stable attachment and synchronized behavioral recording allow activity to be tracked across locations and tasks. Researchers use head-mounted microscopy to measure calcium dynamics in identified neurons, examine circuit responses during learning and social interaction, and relate cellular activity to movement and decision-making. Its portability expands in vivo imaging beyond restrained preparations and supports more ecologically relevant studies of brain function.

2020

In vivo microendoscopic calcium imaging is an invaluable tool that enables real-time monitoring of neuronal activities in freely behaving animals. However, applying this technique to the amygdala has been difficult. This protocol aims to provide a useful guideline for successfully targeting amygdala cells with a miniaturized microscope in mice.

Y-shaped cutting measures fracture-relevant length scales and energies in soft materials. Previous apparatuses were designed for benchtop measurements. This protocol describes the fabrication and use of an apparatus that orients the setup horizontally and provides the fine positioning capabilities necessary for in situ viewing, plus failure quantification, via an optical microscope.

Layered Agar Mounting: Preparing Live Zebrafish Embryos for Long-Term Imaging with an Inverted Microscope

Young children do not passively observe the world, but rather actively explore and engage with their environment. This protocol provides guiding principles and practical recommendations for using head-mounted eye trackers to record infants' and toddlers' dynamic visual environments and visual attention in the context of natural behavior.
