Perfused Mouse Brain

A perfused mouse brain is brain tissue prepared by circulating a physiological solution and often a fixative through the animal’s vascular system to preserve structure for neuroscience research. In transcardial perfusion, fluid enters the heart and passes through cerebral blood vessels, flushing blood before fixation stabilizes cellular and tissue architecture. This preparation supports histology, immunohistochemistry, microscopy, and neuroanatomical analysis by reducing postmortem degradation and improving tissue preservation. Researchers use perfused mouse brains to examine neural circuits, protein distribution, disease-related changes, and responses to experimental treatments with greater structural consistency.

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.

We describe a protocol allowing the purification of the mouse brain's vascular compartment. Isolated brain vessels include endothelial cells linked by tight junctions and surrounded by a continuous basal lamina, pericytes, vascular smooth muscle cells, as well as perivascular astroglial membranes.
