Respiratory Network Analysis

Respiratory network analysis examines how interconnected neurons and brain regions generate, shape, and adapt the breathing rhythm, making it a central approach for studying neural control of respiration. It combines measurements of neuronal activity with circuit mapping and computational analysis to identify interactions among rhythm-generating networks, sensory inputs, and motor pathways that coordinate respiratory muscles. In neuroscience, this framework helps explain how breathing responds to changing physiological demands and how disruptions in respiratory circuits contribute to neurological disease. It also supports the evaluation of experimental models, neural interventions, and potential therapies for impaired breathing.

Here, we describe a protocol to record and analyze respiratory electromyography (EMG) signals. It includes the anatomic references for placing the EMG electrodes over several respiratory muscles, removing electrocardiographic noise from the EMG signals, and acquiring the EMG root mean square (RMS) and onset timing of activity.

The central respiratory drive is located in the brainstem. Spontaneous respiratory motor output from an isolated brainstem-spinal cord is recorded by placing an electrode on the fourth ventral root. This experimental approach is valuable for pharmacological investigations or the assessment of respiratory challenges and genetic manipulations on rhythmic motor behavior.
