Temperature Inducible Expression

Temperature-inducible expression is a gene regulation method in which changing temperature activates or increases production of a selected protein, providing precise temporal control over cellular experiments. Typically, a temperature-sensitive promoter or regulatory system responds to a defined temperature shift by altering transcription, while returning to baseline conditions can reduce or stop expression. In neuroscience, this approach can control fluorescent reporters, signaling proteins, or neural activity modulators in cultured cells, model organisms, and engineered tissues, helping researchers examine developmental timing, circuit function, and cellular responses while limiting unwanted effects from continuous gene expression.

The method described here is used to induce the apoptotic signaling cascade at defined steps in order to dissect the activity of an anti-apoptotic bacterial effector protein. This method can also be used for inducible expression of pro-apoptotic or toxic proteins, or for dissecting interference with other signaling pathways.
